Bill - as we discussed last month, I've spent some time thinking about how to make
progress in my own domain. This presentation represents the first step into a field where
I think there is huge opportunity - a new way to think about neurotherapeutics. The cool
thing is that this therapeutics applies to all organ systems and not just the brain.
1800s were about blood transplants to supplement oxygenation. 1900s were about bone
marrow transplants to augment immunity and organ transplants to recover function. The
2000s will be about organelle transplantation to improve cellular performance and
healthspan. My idea to transplant mitochondria into ischemic brain stems from this belief.
A few things to keep in mind:
1. This is cutting edge science and the field is still small, despite some of the stories I
suspect you have been told. Time to jump in is now. Experiments have been successful in
the brain and they are outlined in this presentation. This isn't crazy science, it's now a
race.
2. I'm proposing this as a three year plan, the first year mostly a series of contracts and
grants - with growth beyond that based on what we learn. I would like to remain in
stealth mode as long as we can and until our IP is secure. We can move faster or slower
based on what we learn.

3. Some of the IP has been described to you, Larry and Casey. Most of it can apply
broadly across all cells - not just the nervous system. This business proposal is meant to
dive deeply into the neuro space as the testing ground for some of the "IP-able" ideas,
since the neuro space is one of the most difficult.
4. I'd like to base this effort in Seattle but due to the very limited expertise, a lot of the
experiments will have to be done outside Seattle and within a few other academic
institutions such as Harvard, MD Anderson and Johns Hopkins. We will be piggybacking
on existing IRBs and animal models but will still need skilled lab techs and credible
experimentation to move this forward.
5. I'm asking you to fund this whole first part (2-3 years, $6 million). This will allow us
to move quickly, leverage some of the existing Gates resources, and work in stealth
mode for a bit. I've spoken some to Bob Nelsen (but not told him very much) @ Arch, the
folks at Third Rock and the Greylock team - and if we can get beyond this first phase we
can all decide together whether we grow alone, with partners or sell.
Beyond the request to fund this I'm asking two things: please keep this confidential for
now / don't circulate and please let me know soon what you think. Science is moving
very, very quickly and I want to take the lead.
And to Larry, Boris, Tony, Philip and of course Lowell - thank you all for your support. I'm
grateful Tony is letting me be Modern Neuron. Comments welcome and appreciated but
mostly I'd like to get started. And stop making slides.
